Preliminary investigation of effects of centrifugation condition on several biochemical tests

Abstract

Objective: To assess effects of centrifugation time and relative centrifugation force in pre-analytical procedure on results of glucose, urea, creatinine, triglyceride, cholesterol, HDL-cholesterol, LDL-cholesterol, aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma glutamyl transferase (GGT), acid uric, total bilirubin, direct bilirubin in plasma samples. Subject and method: 258 whole blood samples of 86 patients examined at Department of Examination - 108 Military Central Hospital from 15/11/2020 to 15/02/2021 that were collected in lithium heparin tubes (3 tubes per patient). All of whole blood tubes were centrifuged on 3 different conditions, 1st tube (at 3000 revolutions for 5 min), 2nd tube (at 3000 revolutions for 10 min) and 3st tube (at 4000 revolutions for 3 min). After that, all of plasma samples were analysed on AU5000 system (Beckman Coulter). Result: There were statistical significant differences between the concentrations of glucose, LDL-cholesterol of samples centrifuged at 4000 revolutions for 3 min compared with those of specimens centrifuged at 3000 revolutions for 5 min and 3000 revolutions for 10 min (p<0.05), with different concentration values were 5.64%, 8.11%, 9.82% and 10.86%, respectively. The concentrations of creatinine and cholesterol of specimens centrifuged at 3000g for 5 min and 4000 revolutions for 3 min differed significantly (p<0.05), with the proportion change in concentrations were 3.09% and 3.0%, respectively. The remaining tests (urea, triglyceride, HDL-cholesterol, AST, ALT, GGT, acid uric, total bilirubin, direct bilirubin) had the similar concentrations on all 3 centrifugation conditions. Conclusion: Blood samples were centrifuged at 3000 revolutions for 5 min without impacts on the results of glucose, urea, creatinine, cholesterol, triglycerid, HDL-cholesterol, LDL-cholesterol, AST, ALT, GGT, acid uric, total bilirubin, direct bilirubin in plasma samples.
