Abstract

Objective: To formulate and characterize phytosome incorporating bioactive constituents extracted from Herba Cistanches (phytosome NTD). Subject and method: The preparation of Phytosome NTD was achieved through a combination of solvent evaporation and lyophilization techniques. The study employed entrapment efficiency, particle size, and polydispersity index (PDI) to examine the influence of various factors, including ethanol concentration, solvent volume, NTD extract/lecithin ratio, reaction temperature, and reaction time. Furthermore, assessments were conducted on morphology, particle size, PDI, zeta potential, FT-IR spectroscopy, differential scanning calorimetry, and the oil–water partition coefficient. Result: Under ultrasound 37 kHz conditions, phytosome NTD was obtained with the following conditions: A ratio of NTD extract/lecithin of 1/1; a reaction solvent of 40% EtOH; a solid/solvent ratio of 1/50; a reaction temperature of 70°C; and a reaction time of 4 hours. Phytosome is spherical in shape, with particle size < 200 nm, polydispersity index < 0.3, and a zeta potential of around -25.77mV, entrapment efficiency of about 75%. Conclusion: Phytosome NTD was successfully formulated, and several of its important physicochemical characteristics were evaluated.
