Genetic diversity of cox1 mitochondrial gene among Opisthorchis viverrini isolated from patients in Phu Yen province

Abstract
Objectives: This study was conducted to analyze the genetic diversity of the mitochondrial cox1 gene of O. viverrini in Phu Yen province, Vietnam. Subject and method: Twenty adult O. viverrini individuals were collected from various patients in An My and An Hoa Hai communes, Tuy An district, Phu Yen Bai. Among them, eleven were sequenced to obtain the complete 1551 bp of cox1 gene sequences. Subsequently, the complete cox1 gene sequences were compared with reference sequences (JF739555.1, China) from GenBank to determine genetic diversity. Results: Fifteen polymorphic sites in the cox1 gene sequences of O. viverrini were identified at positions 15, 150, 152, 236, 321, 324, 354, 384, 407, 585, 738, 1257, 1275, 1305, and 1428. Nucleotide substitution at position 152 resulted in a Threonine → Methionine amino acid substitution, at position 236 in an Isoleucine → Asparagine change, and at position 407 in a Glycine → Alanine change. Conclusion: The mitochondrial cox1 gene sequences exhibit high genetic diversity and play an important role in analyzing the population genetic structure of the small liver fluke.
