Next-generation sequencing for detection of gene variants in non-small cell lung cancer tissue samples

Abstract
In next-generation sequencing (NGS) to detect genetic variants in non-small cell lung cancer (NSCLC) for targeted therapy, it is essential to obtain and validate the results of NGS to ensure the accuracy of the results and prevent false positives or negatives that could impact treatment decisions. We performed NGS to identify genetic variants for targeted therapy on 30 tissue samples from fine needle aspiration and surgical biopsies of NSCLC patients at 103 Military Hospital, utilizing the Ion Torrent system and a DNA and RNA target sequencing kit to assess variants in 81 genes. Validation through Digital PCR and Minisequencing on 30% of the samples showed 100% concordance with NGS results. This study employed a reliable approach to detect genetic variants in NSCLC, laying the foundation for the application of targeted therapy in Vietnam.
