Investigation of single nucleotide polymorphisms in TNF-α gene and their correlation with hematological parameters in psoriatic arthritis patients

Abstract

Objective: To identify the genotype distribution of TNF-a single nucleotide polymorphisms (rs1799964 and rs1799724) and hematological parameters in psoriatic arthritis. Subject and method: The case-control study was conducted at Ho Chi Minh City Hospital of Dermato-Venereology (from March 023 to November 2023) including 43 psoriatic arthritis (PsA) and 43 psoriasis vulgaris (PsO) matched by age group and gender. We confirmed the PsA diagnosis based on CASPAR criteria. Hematological values were recorded from hospital’s results; TNF-a single nucleotide polymorphisms (rs1799964 and rs1799724) were detected by Sanger sequencing method. Data analyzing was performed by SPSS 25. Result: The mean value of disease duration of the psoriatic arthritis group was statistically significantly higher than that of the plaque psoriasis group (p=0.035). The rate of the TT genotype of rs1799964 in the psoriatic arthritis group was higher than the plaque psoriasis group and vice versa for the CC genotype of rs1799724, however the differences were not statistically significant. In psoriatic arthritis patients carrying the T allele of rs1799964, the NLR index was statistically significantly higher than the group carrying the C allele. Conclusion: Except for disease duration, there were no statistically significant differences in genotype distribution of two single nucleotide polymorphisms (rs1799964 and rs1799724) between the two groups. Moreover, a statistically significant association was noted between the allele type of the single nucleotide polymorphism rs1799964 and the NLR index in the group of patients with psoriatic arthritis.
